What Is Escin?

Escin is a naturally occurring mixture of triterpene saponins extracted from the seeds of Aesculus hippocastanum, the horse chestnut tree. Used for centuries in European botanical medicine for its anti-inflammatory and vascular-protective properties, escin is among the more extensively studied botanical compounds in the literature. Its molecular class, oleanane-type pentacyclic triterpenoid saponin, allows it to interact with several biological pathways.

Mechanism

What Does Escin Do?

Escin exerts a glucocorticoid-like anti-inflammatory effect by upregulating the glucocorticoid receptor, suppressing NF-κB signalling without increasing steroid hormone levels. It reduces vascular permeability by protecting PECAM-1 distribution in blood vessel walls, limiting fluid leakage into surrounding tissue. It inhibits hyaluronidase, the enzyme that degrades hyaluronic acid. And horse chestnut extract standardised to 70 percent escin scavenged free radicals above vitamin E in an in vitro screen. These pathways are documented in laboratory and animal studies.

Glucocorticoid-Like Anti-Inflammatory

Escin upregulates the glucocorticoid receptor, which suppresses NF-κB signalling, the master switch for inflammatory gene expression. In laboratory and animal studies this produced an anti-inflammatory effect comparable to steroids without increasing glucocorticoid hormone secretion. Applied externally in animal models, escin reduced prostaglandin E2, TNF-α and IL-1β in skin tissue. The pathway is well documented in vitro and in animals, and has not yet been confirmed on human facial skin.

Mechanism II

Vascular Protection & Barrier Repair

Escin prevents hypoxia-induced disruption of PECAM-1 distribution, helping protect blood vessel wall integrity and limiting fluid leakage into surrounding tissue. This is the mechanism behind its anti-oedematous effect, studied in chronic venous insufficiency, where one trial suggested oral horse chestnut seed extract may be as effective as compression stockings. The vascular mechanism is documented; a topical effect on facial skin has not been demonstrated in a clinical trial.

Mechanism III

Hyaluronidase Inhibition & Antioxidant Defense

Escin inhibits hyaluronidase, the enzyme responsible for degrading hyaluronic acid in connective tissue. In a formula containing hyaluronic acid this suggests escin may help protect the applied HA from enzymatic breakdown, though direct confirmation in human skin has not been identified. Horse chestnut extract standardised to 70 percent escin also ranked among the highest active-oxygen scavengers of 65 plant extracts tested in vitro, above vitamin E.

Why Escin for Skin?

No competing skincare brand positions escin as a hero ingredient, despite a Cochrane review pooling 17 randomised trials of the oral extract. In animal models escin produced anti-inflammatory activity comparable to a steroid, without immunosuppression. Its documented mechanisms, reduced vascular permeability and hyaluronidase inhibition, are why we chose it as our foundational active and pair it with hyaluronic acid. Trials on human facial skin have not yet been run.

A Cochrane systematic review pooled 17 randomised controlled trials of oral horse chestnut seed extract in chronic venous insufficiency, finding significant anti-oedematous and symptom benefit against placebo. That evidence is for the oral extract on a vascular endpoint, not for topical use on facial skin.

No Immunosuppression

Observed In Animal Studies

In a mouse model escin's anti-inflammatory effect lasted longer than dexamethasone, with no immunosuppression observed. No safety concerns have been identified in peer-reviewed literature at cosmetic concentrations of 0.5 to 2 percent.
